What Lion’s Mane Mushroom Is

Two groups of compounds carry the marketing. Hericenones sit in the fruiting body, the part that looks like a mushroom. Erinacines sit in the mycelium. That is the root-like web underneath, and it is grown in a lab rather than picked.

The Nerve Growth Story

The hype has a real idea behind it. The usual explanation is nerve growth factor, a protein that helps nerve cells survive and repair. What The Human Trials Show

Mori 2009 is the trial everyone quotes. Thirty Japanese adults aged 50 to 80 took part, all with mild memory problems. They took 3 g of dried fruiting-body powder a day for 16 weeks (Trial). Then comes the detail almost nobody quotes. Four weeks after people stopped taking it, their scores dropped again (Trial). That suggests the mushroom was propping the scores up rather than rebuilding anything. Take it away and the effect goes with it.

A later trial team read it the same way. They noted the gains were gone at the check four weeks after treatment ended (Trial). An independent expert review adds a second caveat. Saitsu 2019 tested 31 healthy adults over 50 for 12 weeks. One test out of three improved. The same test also improved in the dummy group (Review).

Li 2020 is the longest trial anyone has run. It was a 49-week pilot in people with mild Alzheimer’s disease. A coin flip sorted 49 people into two groups, and 41 finished (Trial). They took about 1 g a day of mycelium rich in erinacine.

Scores on a common 30-point thinking test rose by about one and a half points in the treated group. That was a gain on their own starting point, not a win over the dummy capsule. Docherty 2023 is the most careful healthy-adult trial in the set, and its results are humbling. A coin flip sorted 43 young adults, and 41 finished 28 days at 1.8 g a day (Trial). One hour after a single dose they were faster on a color-naming attention test, by about a twentieth of a second (Trial). The authors call that a one-task result in a small pilot, and say to read it with caution.

Surendran 2025 is the newest published trial and the smallest. Eighteen healthy adults took one 3 g dose of a fruiting-body extract of set strength. They sat tests 90 minutes later (Trial). Overall thinking and mood scores did not move.

Task by task, the results pointed both ways. Hand skill improved. Two attention tasks got worse, and the rest did not shift on lion’s mane (Trial). The extract was well described, so a cheap jar is not the excuse here (Trial). Two of the authors work for a company that sells lion’s mane drinks, and that company helped pay the publishing fee (Trial).

One more study is doing the rounds in 2026. A coin flip put 150 adults aged 40 to 75 into two groups for eight weeks. One group took a blend of fruiting body and mycelium, the other a dummy. Of the 109 counted in the results, 57 took the mushroom, and one attention task improved more than the dummy (Preprint). It is the largest trial yet. One gap is bigger than any single trial. Nobody has pooled these trials into one answer. Is Lion’s Mane Safe
